Yes it should. I have a different board, Gigabyte GA-Z170XP-SLI. 
You just need to look for '288 pin', '3200', 'DDR4'. You should be good. That's a sick board!

So, I have a situation I hope someone out there can sort out.  I just bought a 2nd set of these RAM sets to populate the remaining slots of my mobo and my computer is constantly crashing now that they're installed.  Anyone know why?  I've already confirmed that it's not the Ram sticks causing it.

You can try turning your fan speeds up. I had mine on silent and the same thing kept happing to me. I am assuming you know your uefi settings or you might know it as your bios. Standard setting worked for me.

I don't know exact height but they weren't tall at all. I had enough clearance to put a BeQuiet air cooler on Threadripper

This RAM works fine on my ASRock Fatal1ty Z270 GAMING K4 LGA 1151 Intel Z270 HDMI SATA 6Gb/s USB 3. motherboard. Its DDR4 memory standard RAM and I'm sure should be compatible with Asus Z370-E motherboard.  I would buy this RAM again.

If I have 2 of my 4 slots filled with 8 GB sticks can I fill the other two slots with two 16 GB sticks or do all 4 slots have to be the same size/speed?  I have the ASUS PRIME X570-P ATX motherboard.

I would stay with the same brand and speed but you can install a pair of different size memory as long as the PAIR is in the correct slots as indicated by the manual. It is usually one pair would be in the A1 and B1 slots and the second pair would be in the A2 and B2 slots. They work great with z270 boards.   Most cpu cooler come with something to raise for ram slots and these are very low profile
