is this usb card compatible with a mobo that has  these expansion slots open -PCIe 2.0 x16 , PCIe x1 and PCI?

Yes, but only the 16x pcie.  It is a pcie x4 card

I don't have that exact combination but the card should be compatible with any motherboard with an available PCIe x8 slot. If it works and works well in my old ASUS 1366 board with PCIe 2.0, I can't imagine why it wouldn't work in the latest editions. As with anything, it may be necessary to find the right driver for it. I don't recall having any issues installing the card and loading drivers included with it.

It is amazing what a difference USB 3.x has on transfer rates. Probably now only takes a 1/10th of the time it used to take to image a hard drive using a 2.0 interface. Something like going from 15-20 minutes down to 2-3 minutes.
