Q: Do are both molex cables required to be plugged in, or only one?

both, if you're trying to convert a six pin into 8pin for a new GPY that have a six and 8pin, u have to get the molex dual 4to8pin and both molex have to be connected to the PSU, since the new GPU requires extra power

Can the 8 pin be split into a 6+2 pin?

This adapter unfortunately cannot be split into a 6+2. It's a single connector.

Question 2

Can this adapter work for a 8 pin connection on a motherboard as well? Or does this 8 pin only power PCI-E Graphics cards. It's not very clear on the description.

There is a difference in pin-out between a PCI-E and a EPS connector, both with power distribution and with the "Rounded/squared peg" configuration. So no, this would not work as an 8-pin CPU power plug.

I'm looking at building a rig with a Sapphire R9 280X that requires "2 150W 8-pin PCI Express power cables" and I hadn't been aware I would need to buy cables separately, are these what I want?  AMD Build w/ Gigabyte MoBo if that info is needed to answer.  THANKS MUCH.

Yes this is the same 8-pin cable that you need for that video card. It sounds like you need two. However, the video card may have come with an adapter similar to this, and your power supply should have at least one of these type of connectors on it already - if not two of them already. I would check that before ordering these.

How much power can these carry?  Is there any chance of damaging my GPU (assuming my PSU has the wattage needed for the cards)?

It's only a few inches of wire on this, so the answer to your question is different for each particular PSU. Best chance of success is to join each of these two white ends to a different "daily chain" emerging from the PSU metal shell. And, if possible, >NOT< shared with other drives on the same chain...drives "brown out" the full 12Volts momentarily as their electromagnetic coils kick in pulling big slugs of current.
