I have just been given a computer system with this cooling system installed. It's been sitting in storage for three years. I've never used a liquid cooling system for a computer before. What should I do to make sure it's all ready to go?

I built a computer with this cooler about 1-1/2 years ago and have used it nearly every day without an problems. The cooling unit is factory-sealed, and there is no maintenance required by the user. If you're concerned about leaking - especially if you don't know the history of your computer - it might be a good idea to operate the computer with the side panel removed until you are confident that the seals are tight. Also, monitor the temperature of various components, either with the Kuhler software, the Motherboard's software, or similar utility.

My Khuler H2O 920 HAS BEEN WORKING GREAT FOR 3YRS NOW
and one day recently the temp. started to rise slowly from 38 deg.C to 51deg.C over a 20 minute period and the fans going almost full speed.

What should I check for first?

Have you eliminated high-CPU activity?  If so: Check for dust, debris in the fans.  Check for a stuck fan or poor connection.  Most of all check for evidence of fluid leak around the connectors.
