Will the displayport use the garphics card or the motherboard?

It connects to the TB3 (Thunderbolt 3) header on your motherboard. Because TB3 is tech owned by Intel, very few AMD MBs have the header...do your homework to make sure. You also connect a DP (mini display port) in from your GPU, to then transmit out an up-to 8k picture by the TB3 ports on the back. Hope this helps.

Will this work with AMD boards, more specifically a B450?

I'm successfully using it on an AMD ASRock X570 Taichi board. If your board has the thunderbolt connectors on it then there's a good chance it will work as the drivers are pretty generic Intel ones, but i'd check your motherboard support docs for what they say to be sure as I previously used a Gigabyte Titan Ridge on my ASRock X570 and while it worked there was a quirk with the computer not going to sleep if I left a thunderbolt device on, this resolved when I used the ASRock TB card.
