How does this hook up to my router?

Depends: If your router has an external antenna (or antennas), just unscrew one of them, and attach the asus one in it's place.  However, some routers have external antennas that are fixed; that is, they are not removeable. - in that case, since the antennas are not removeable, you can't attach the ASUS one.  And, if your router only has internal antennas, you can't attach the asus one.

Anyone notice an increase in their 5GHz frequency?

Will this fit on the ASUS RT AC66R?

I would imagine it would be compatible. Seems like a standard size connector. That router looks like my RT-N12 that I have this antenna plugged in to.

will this plug in to my verizon jetpack or at least compatable ?

It will not plug into jetpack. It has coax connection.
