How does this actually mount to the desktop? Is is a clamp that can be removed (Hopefully) without any perminent marks? Or does it have to be bolted down?

It actually has both options (bolt and clamp). I ended up clamping it to my desk and it is pretty sturdy. There should not be any damage to your desktop unless you scratch it during the install. Hope this helps.

There aren't any "locks" to set. You can tighten things up so that they don't move except under higher pressure. You use an allen wrench or screw driver to do this.

Can this come for a single monitor?

I think that it would be some what off balance but after looking at mine, I think that you could only use one monitor.
