Should M.2 drives have heat sinks for this MOBO or not?

If you are using a m.2 drive that is utilizing PCIe 4 you will need a separate heat sink if you want to reach the max read/write speeds.  If you're using a PCIe 3 m.2 drive then the integrated heat sinks are fine.

The 3 M.2 slots are not the same?

All slots are m2 Nvme capable. And can accept 2280 type SSDs. However one of them is connected to the TRX40 chipset sharing its PCIe lines, while the other two goes directly to the CPU. This difference has the potential to translate on somewhat lower performance for the TRX40 port, but I guess, in practice, the performance should be OK as most SSD can't saturate a PCIe 4.0 port.
