Does this card have a driver for Windows Server 2010 R2?

I cannot answer this question from personal experience, but I will tell you the cards are plug and play with no driver installation under Thecus NAS OS as well as VMware ESXi 5.x. I would be a paycheck or two that it's supported under Server 2008 and Server 2012.

Is this compatible with the newer MOTU avb/thunderbolt audio interfaces?

I cannot answer as I don't have a newer MOTU avb/thunderbolt audio interface. It did solve the problem of my desktop class motherboard internal NIC being incompatible with Windows Server 2012 R2. I even tried hacking the driver for the desktop NIC and I still ran into random crashes. This solved the problem completely.
