Does this work with just one 2.5" SSD?

Yes, for installing a single 2.5" SSD unit into a 5.25" standard CD-ROM bay.  The additional height is of no consequence, as it will fit perfectly in a 5.25" drive bay with your SSD unit installed in it.

There may be other solutions to use two SSD units in a single drive bay, but I prefer having more air flow between the hardware, just my personal preference!

No.  As stated in description, it is for mounting up to two 2.5 inch hard drives in a 3.5 inch slot.  Each side of unit is 1/2 inch wide so reduces width by 1 inch (not 1.75 required for 3.5 to 5.25).

As a side, unit comes with 4 screws to mount to exiting 3.5" slot and 8 screws to mount the 2.5 inch drives.
