The specs do not indicate that it is Blu-Ray so you should assume that it is NOT Blu-Ray

I'm not a 100% sure but I think it does support it. Most modern gaming laptops do.

The M.2 SSD interface is PCIe. (A SATA M.2 SSD will not work.)

I am not sure that I am qualified to answer this questions.  It has a webcam, so I m sure it has a microphone; but I can not comment on the quality.  I am sorry...

I do very intensive video editing with lots of vfx and layers, and need flawless timeline playback with no lag issues. can this unit handle it?

I edit weddings with multiple tracks, 1080P 60FPS, and the time line has no lag whatsoever.  I use power director 14 and keep the preview on HD, but not full HD and it previews effortlessly.   I tried the 6500 new i7, and it wasn't powerful enough.  This baby is awesone.  Suggestions: put at least 32gb or ram, and order the country with a bigger ssd drive.  The OS comes on the 128 gb, and it's not enough,  but is wicked fast.  I can render a full HD, 60FPS video 4 hours in length in under 4 hours.   It would have taken 8 times that on an AMD A10 with 24gb of ram. Best tech purchase I have ever made.

is it possible to upgrade the graphics card to a 1060?

I honestly don't know for sure but I would not think so because you would need a 1060M card and I have not seen one for sale anywhere (not that I have really looked). Also, I have not accessed the interior of the laptop in quite awhile but I do not remember seeing a way to access the video card from the access plate on the back of the laptop (but I am sure it could be done somehow). You would be better served by asking Asus customer support if it is possible, they would be the ones to know for sure.
