I have a Rosewill HDD enclosure and it has an ESATA port at the backside of the enclosure. I don't have any ESATA ports. I need to know if this plug are both female connectors. I need it to be female ESATA to female SATA. The ESATA female end for it to plug into the enclosure and the other end female SATA, so that that i can plug it into the SATA port found at the backside of my PC. Are they both female?

You most likely need an Esata to Esata. I've never come across a MB with a sata port on the back of the board. Sata is generally internal where as Esata stands for External Sata, hence it is likely the port on the back of your PC.
