Will this work with a SATA III port on the motherboard and a SATA II optical drive?

Yes. The transfer speed will be cut in half due to the optical drive being SATA 2. The cable connections are the same. The difference is the transfer rate. SATA 3 is about twice as fast as SATA 2. It is like USB 2.0 and USB 3.0.
