I want to connect a 2014 mac air (Thunderbolt connection) to my 2010 imac (no thunderbolt). Will this lead do the job?

Yeah, you need this cable because although the thunderbolt port on the Macbook Air should be reverse compatible with Mini DisplayPort, the iMac won't recognize the Mini DisplayPort signal coming through a thunderbolt cable.  Both computers will be able to use this cable though (only for display no data transmission).
