Will this work on pci express x1?

no it is a X4 slot compatible although it can use pci-e 2.0 slot like most motherboards have one X16 pci-e 3.0 and one X16 pci-e 2.0 use that slot if you have it free

I would just like to add that my system - MSI X370, Ryzen1700X (OC 4.1 GHz), 32gb GSkill DDR4 @2800, Radeon RX 480, and OS - WIN 10 PRO X64) - is completely UASP compliant. While the price of this card is very inexpensive and its true there is no direct mention of it being UASP compliant on its own site or anywhere else I looked, I definitely got the feeling it was implied or meant to be "understood" in more than a few different descriptions that I read about this card. Finally, the speeds I am getting from it firmly leads me to believe that it is, in fact, UASP compliant and therefore a truly excellent bargain for the price.

Does this card have to plug into a header connection on motherboard, or is that only for the faster 20G-bps & 40-Gbps Type-C Thunderbolt port addIn cards?

It needed a power connection but does not need to plug into a header. Other than the C port being so close to the edge (being elevated a few millimeters would help immensely), it was a snap to put in and use. Being close to the edge makes plugging into the port so close to the case slot edge challenging. Didn't even have to install any drivers.

Does this card support Display Port alternate mode for sending video through a USB C cable? This configuration is used for connecting a Cintiq Pro pen display. Thanks
