Is this a single 16GB stick of memory or 4 separate sticks?

It is a single 16GB stick of memory, I put 4 of them in my workstation for a total of 64GB of system memory.  After using it for almost a year I would highly recommend it.

Is this memory compatible with the new Asus Z170-A ATX LGA1151 Motherboard? I am trying to max out its memory capacity to 64 but I have been unlucky finding 16gb memory sticks compatible with that motherboard.

I've not personally tested it but it is highly unlikely to work.  This is ECC Registered memory.  The Asus Z170 lineup are desktop boards with desktop processors that do not support ECC memory, nor registered memory.  Only non-ECC un-buffered (AKA un-registered) memory is supported.  It's unlikely that ECC would be an issue but the buffered is for a couple reasons.  Registered/buffered memory has higher latency.

will this be a drop in replacement for "Samsung 8GB DDR4 Desktop Memory M378A1K43CB2-CRC RAM" ?

No, the 16GB module is RDIMM meaning it is registered and your other module is UDIMM which is unbuffered.
These have differing applications and should not be mixed.  If your motherboard supports registered ecc ram then they is the way to go, otherwise stick with udimms.
