Is this backward compatible with USB 2.0?

Yes, it will work fine on USB 2.0.  Just not as fast as SB 3.0 obviously.  I run mine with an Intel 256GB SSD drive and  works great on either 2.0 or 3.0

Does this device support 1TB 2.5" SATA III Drives?
I own the "HGST 1TB Travelstar 2.5" Mobile Hard Drive"
B&H # HGHDIS25 MFR # 0S03563 which is essentially a 2.5" internal SATA III driver. I would have purchased this drive from NewEgg, but B&H have always been cheaper than all other online stores, for this specific drive which is an AWESOME Laptop Drive.

The internal connection fits a standard SATA power and data plug, not slim or mini.  This is the same as all of the other SATA HDDs in my collection, (except for one mini in a laptop that is clearly different.)

Not sure about your specific application.  I did swap a few SSDs and HDDs through this during a small build and it worked with all of them.  I formatted a few drives with it before I put them in the case.  The little board seems to be a simple, effective interface to USB3.0.

The case is only big enough for a naked chips-on-PCB style drive.  If you have something bigger you probably need to look for something else.
