Do I have to have the box powered on and connected 24/7 to use my computer? Or, can I disconnect it from my PC and only hook up the cable/external box when I want to use it?

I don't think it could be compatible with a laptop. I am not sure why you would be thinking of this with a laptop because it's a PCIe to PCI bus interface.  This is for a desktop motherboard where you need PCI slots and you only have PCIe.
