Does  work on my Asus motherboard with Win 10.

Be SURE you are connecting these cards to a PCI 2.0 slot (and not the small 1.1 slot on your motherboard). I looked up your motherboard and you have 3: PCIe 2.0 slots, 1: PCIe 1.1, and a PCI slot.

Bandwidth** (you would not have 5Gbps on a diminutive 1.1 slot). ie: same slot as a video card would use if that helps you. Also note that ESATA cards use the same slots as well.

My computer has a Gigabyte GA-MA785GM-US2H Quad-Core mainboard that I would like to upgrade to USB3 capable.  The board has one (1) PCI Express X16 expansion slot and one PCI Express X 1 expansion slot.

Can the StarTech 2 Port PCIe (PEXUSB3S25) adaptor be used with the PCIEX1 slot?

That should work fine, as the PCIe lanes are designed to be compatible with shorter cards. For example, a graphics card is normally a PCIe x16 lane and can operate cards that are PCIe x16,x8,x4,x2, and x1 widths.

Just be sure to insert the card straight and secured to the back of your case so you do not inadvertently short something out.
