why is there no 2280 PCIe M.2 NVMe SSD adapers to a sata 2.5 slot out there?

NVMe chips are closely related to PCI-E components, but SETA chips are related to SETA hard drives. Though they both store data and use a similar physical connector, these are completely different classes of device, and such a converter would still involve a ribbon cable going from your 2.5" mount to a PCI-E slot, which would typically be the opposite side of the computer case. Since M.2 SSDs are incredibly small and lightweight compared to hard drives, there's no reason to go through extra trouble and mount it in the 2.5" slot, which is designed to hold really bulky frames. Instead, they just get mounted directly to the motherboard or on a PCI-E card.

Question 2

Can this be used to create a bootable system?  First drive with OS and second drive with storage (Drive C & Drive D)?  My PC only has one SATA connector for a hard drive (All in one PC).

It can be a bootable drive but it has to be a RAID configuration, you have to see if that desktop supports RAID. I dont believe it will work as you listed.  Refer to the various types of RAID defintions 0,5,10.  If your pc supports any of those you can achieve what you want.

this B key M.SATA only devices? what about M key, will it fit 2280 length?

This device supports B key M.2 SATA SSDs and is not compatible with M.2 NVMe or AHCI PCI-Express SSDs. It supports full-length M.2 SSD drives of 2280 size.

Question 4

I can't imagine the scenario where this would need to exist.  Wouldn't a PCI-e to b-key M.2 make more sense?

It would depend on how many slots you have, if any.. I used this in my laptop to combine 2 smaller M.2 drives into one and used my SATA drive
slot, then replaced my one slot with a larger m.2 drive, thus giving me the space boost and performance boost I needed.. I have no physical HD in my laptop anymore.

Question 5

Do you have to run these in a RAID or can you access each drive separately still?

Stripe, Mirror and Span should work fine as these modes are handled locally on the device itself and appears as a single drive to the system. JBOD mode allows for each drive to be seen individually ONLY IF the SATA host controller it connects to has support for the Port Multiplier Protocol. If PM support is not featured then only one M.2 card could be seen by the OS. FYI - No Apple machines have PM support on their standard connector.
