Do the 16GB (2x8GB) 1600MHz, DDR3, 240-pin memory kit like this G.Skill run on the ASUS M5A99FX Pro motherboard? Would two og these (total of 32GB) work. I plan in install Win 8.1, 64-bit.

unless you are building a CAD station or some other highly graphic intensive purpose, in which case you should focus on the ram of the needed graphics card, 32gb is going way overboard. I have 16gb and only because 8 were free... I run multiple virtual machines and i never crack 50% usage.

Is the speed on this memory 1600 SPD, or do I need a PC that can enable XMP (which mine does not)? In other words if it needs a system that can enable XMP to get 1600 speed, my system will down clock it to 1333 - which I don't want.

It's 1600.  XMP is a method of automatically overclocking which means it drives things above the rated speed.  This ram does support XMP if your system supports it.  If you find your system defaulting to some lower speed, you can go into your bios and pick a higher speed profile.  If you're bios doesn't support memory profiles then technically it shouldn't adjust timings, latency, or voltage either ... but you never know.
