Question 1

My computer has 6 gb of ram-3 x 2gb with one open slot.  Is there any reason I can't use this 4gb in the one open slot?

Short Answer: You Shouldn't do that

Long Answer: When picking ram you should always buy and plan to upgrade in pairs. that being said when buying new ram to add to existing system you need to be aware of every technical features of the existing ram, including latancy, speed, ddr 2 or ddr3 and etc. On top of that Windows more than likely wont recognize the extra 4 gigs since it is not setup in dual chaneel on your MB

What rank is this memory. I've been advised 1R is better than 2R for my Dell Inspiron 620 desktop?

I'm not sure about rankings. This ram has worked well for me since I bought it which was for the price. My computers are used daily and never an issue. Hope this helps

This memory will work in pretty much any DDR3 motherboard that is rated for at least 1333. My experience is that MB's will clock down to slower RAM, but faster RAM will not always work in slower boards.

Even though you can, you shouldn't mix memory.
They should have the same cas latency, timings and voltage (unless you want to modify your BIOS settings).
If the speeds were different then the motherboard will just slow the faster one down to the slower ones speed (not an issue in this case).
Using different sizes will change your computer to run at single channel rather than dual channel, however you as a human probably wouldn't notice the difference.
I would say just buy two of the same at the same time.
If your current operating system is a 32-bit Windows OS then just get a 2x2GB configuration since you cant really use more than 4GB anyway.
