Does it only come with one?

I ordered this ram recently and had to order a quantity of two to get 8gb.   I will be running windows 7 64bit, which will support more than 4gb of memory.  If you look in wikipedia, you will find what versions of windows will run only 4gb of memory and which versions will allow more than 4gb.  Also, consult your motherboard manual to see if you should have two memory cards for best performance.

purchased this memory for HP 4000 desktop. Pulled the current stick and matched up up everything 4GB, PC3, 240Pin, 1.5 V, DDR3-1333, DIMM but doesn't work. Likely reason?

Since it's not possible to put it in backward, I assume it goes in the slot no problem and you are able to lock it in properly. I suggest testing with your old memory in. If everything is fine with the old one, then the new one has a flaw in the circuit board or chip. If things still don't work with the old one, then either the memory slot or the motherboard are corrupted somehow.

Will this work with a dell optiplex 380?

You probably won't be able to use a single 4GB chip.  The spec sheet shows that your max memory is 4 across 2 dims.  That usually suggests that each port will only take a 2GB chip max.  I've had this problem before with an older HP dc5700.  The machine would take 8 GB, but each port took ONLY 2GB sticks.

This is from the spec sheet - Your computer supports a maximum of 4 GB of memory when you use two 2-GB DIMMs. Memory modules should be installed in pairs of matched memory size, speed, and technology. If the memory modules are not installed in matched pairs, the computer will continue to operate, but with a slight reduction in performance.

What I experienced when putting a single 4 GB DIMM in the slot was the PC doing a triple memory beep and failing to start.

It looks like it should work fine though.  The motherboard is requiring DDR3-1066, this memory is DDR3-1333, which is faster, but usually can run slower to meet your need.

So yes, this RAM will be compatible with that board.

Yes, absolutely. If you look at the board here on newegg, you'll notice in the specifications that it accepts 240 pin DDR3 1333 RAM. This will work with your board.
