Does this card still support 4 m.2 drives in a PCI Express x8 Slot or will this limit the card to two drives? I am planning to put it in the fourth slot of my asus rampage vi extreme mb (x16, x0, x16, x8). I have graphics cards in the x16 slots.

If you place this in the x8 slot on your Asus R6E, it will only read the nVME drives located in slot 1 and 2. It will not read/connect anything in slots 3 and 4. I had issues detecting all four drives in the x16 slot on my R6E so be wary that it's not a fully fleshed out product. Currently using 2 nvme drives on the Asus Hyper x16 card + 1 nvme drive in the dimm.2.1 slot (as the dimm.2.2 slot runs off PCH not CPU)  + 1 nvme drive in an Asus Hyper x4 expansion card in order to achieve a CPU managed RAID 0 with 4 nvme drives. The card comes with no software.  Assuming you don't have a Asujs MB per their requirement, And if you're lucky enough to have a recent server MB, you can set your PCIe x16 slot to 4x4x4x4 - usually server motherboards have this option - if they are fairly recent, then it will show up as 4 different drives.   With no ASUS MB that meets their specs, if you can't set your PCIex16 slot to 4x4x4x4 it will just show the first drive.

I was able to set my supermicro MB to 4x4x4x4 on a PCIe x16 slot, and everything showed up and worked great.  Another cheaper server MB with a bios last updated in 2015 didn't have this option.

Are there any AM4 motherboards that this can work on?  I see that it just requires pcie bifurcation.  Plan on putting this card in slot 1 and using x8 PCIe slot for graphics.

No - You need a different card with the Bifurcation Switch Built-In - 2 Options: 
1.) High Point SSD7101A-1 4X Dedicated 32Gbps M.2 Ports to PCIe 3.0 x16 RAID Controller

Your Motherboard has to support PCIe Bifurcation - Meaning - The ability, in the Bios / UEFI firmware to change a X16 Slot to X4X4X4X4. Otherwise, You need a Card that has the PCIe Bifurcation Switch Built-In. Hope that helps!
