No.  Z270 is designed for Kaby Lake, which I'm pretty sure does not support DDR3.  DDR4 is so mainstream now there isnt much need to make new boards compatible.

Can you use the Cooler Master Hyper 212 EVO, CPU cooler, with this board?

Absolutely. The Hyper 212 EVO is keyed for specific CPU Sockets, LGA 1151 being one of them. It's probably more about your case having enough width to fit.

Yes indeed. This board has two m.2 connectors.  One supports both SATA and PCIE mode, while the second  is only for PCIE (that is, NVMe) connection.

I want to have 32GB in my computer, with this mother board, do I need to get 4x8gb?

You can do that and run two dual channel or you could get two 16GB sticks for one dual channel and have two open slots. Just remember that if you wanted to do the later and add more later, it is important that they are matched memory. The memory as a whole will only run at the speed of the lowest stick.  And of course, I am sure you know, but you have to be running Windows 64 bit. Win 32 will not utilize anything over 3-4 GB RAM.

Does the board have front panel connectors for USB 3.1? Asus's specs say no, but newegg's says yes.

No. The only USB 3.1 connections are provided on the rear I/O connection block. One connection is a Type-A port and the other is a Type-C. You could connect either of those ports to a stand-alone desktop hub if you wanted to avoid fiddling with connections on the back of your case.
There is a USB 3.0 connection block on the motherboard that can be used for front panel devices. I am using that with a SYBA USB 3.0 Multi-IO panel and it is working well.

I need to find a case that this will fit into.  Any suggestions?

Like all ATX motherboards, it should fit into any ATX case. Any problems are likely to be stuff that is attached to it, like the CPU cooler. I have a roomy Antec Three hundred Two case that fits this motherboard and Cooler Master Hyper 212 LED cooler no problem. You need a wide case for tall coolers, and the depth of the case might affect how long a video card you can install. Those are the dimensions you need to consider.

That is an LGA1151 CPU and board so compatibility shouldn't be a problem.  I have been running that same board non-stop with an i7-7700K without issues for about 6 months.  Hope that helps.
