I have 2011 v3 X99 ASUS board, will this work with it?

Yes. Even though its a PCIe 4.0 card, and your motherboard is PCIe 3.0, it won't make any real difference to the bandwidth for that card. But you can get relatively cheap motherboards with PCIe 4.0 compatibility now which would also allow you to use M.2@4x4 which are really cheap at the moment and a super fast storage option for gaming.
