I've been wearing these headphones with glasses for hours at a time for years.  There's a minor discomfort with my left ear that I usually forget about until a couple of hours later.  It just might be the shape of my ears, though.  I would say I have medium-sized ears.  The glasses don't really affect the comfort since you they'll stay on if you hook them loosely on your ears.

So in summary, the glasses don't really matter, and these headphones are comfortable enough that I forgot about wearing them for hours..
