Question 1

I'm confused here where does the 2.5 drive mount? It the screw holes are facing up. Can someone let me know where I can find a photo of this with a drive mounted, thanks!

The 2.5 drive mounts inside the bracket using the pre drilled holes on the bottom by the large oval air holes, The bracket then mounts inside any 5.25 bay in your PC case using the pre drilled holes on the side fins.
The 2.5 drive should have the connection sockets at the cut away at the rear of the bracket. (those oval cutouts face down) just like in the illustration. 
Your 2.5 drive will have screws or holes on the bottom.

Will this work in an all-in-one PC? More specifically, the HP Touchsmart 600-1120. Thanks in advance.

If this "all-in-one" PC has a standard 3.5" Hard Drive bay available, then yes, It will work.  All this bracket is doing is allowing a 2.5" (laptop sized) hard drive fit into a 3.5" hard drive space.  The thickness is slightly thinner than the standard 3.5" inch drives, even with this bracket, but that only frees up more space.

Question 3

Will this be suitable for an SSD to be mounted into a hard drive bay specifically in a Dell Vostro 260 Desktop? This desktop has rails that it needs to be attached to and then slid the rails slide into a slot, and is then fastened in place with a screw on the top.

This should be fine for that application. In effect the "Bracket" ends up being the EXACT same dimensions a 3.5" hard drive would be and the holes are EXACTLY where they would  be on a drive. Rails should attach fine. and the 'screws' that hold it in are actually holding the RAILS in, so that should be no problem. SSD compared to an HDD is awesome, go for it you won't be disappointed. Keep in mind an SSD is so lite and so small you might be able to just tie wrap it to the case somewhere and be done with it. I have done that in a few case myself.

Question 4

Can I use this bracket to mount a 3.5" drive to my new Acer box that has mounting holes for 2.5" drives?  I've already added an SSD as the main drive and plan to use the 3.5 as a second drive for data.  2.5 HDDs seem too pricey vs 3.5s.

This is for mounting a 2.5" hard drive in a 3.5" bay.  If you don't have a 3.5" bay then you can't mount a 3.5" drive, however there are options to mount a 3.5" drive in a 5.2" CD/DVD drive bay if you have a spare one of those.
