Does this item require any kind of batteries?

No these don't require batteries because these headphones are neither noise cancelling or active amplification.

Will the headband mess up my wife's  hair-do ?

The band is padded and soft, but unless your wife has a pretty small head, there's no way she could wear them without the band coming in contact with her hair.

That said, 'hair-do' is a little vague, and 'mess up' gives no understanding of degree. If she always has an 'up' do piled on top of her head, and it takes pins and sprays and 30-45 minutes to get right, I'm inclined to say "yes." If the styles she prefers tend toward a flat or slick crown, it should be fine.
