Does this device allow you to "hot plug" disks into its ports?

I did purchase this card, as a replacement for the one that failed.  I'm the end user ~ but when one of the disks failed, my technical advisor explained to me that NO, it is not possible to replace my disk 'hot'...
My RAID 0 {zero} might be part of the problem but I don't think so.
I did not have a spare disk on the shelf, so I had to send the in-warranty one back and wait for the replacement.  Then the tower "rebuilt itself" which took about a week.  
This tower is a backup for my PC.  4 disks of 2TB giving me 3TB from RAID0.  Note, the main file system is similar but 'live' connected to the motherboard, which runs on two Solid disks.

I'm a 70 year old buzzard and just trying to help, but that's what I understand.  My technical expert says 'if you want to upgrade and get to hot-swap, get a Drobo.'

Will this card support all 5 drives on an external bay using just one cable? I have tried with another card that only sees 4.

Yes.  This is actually the card that Sans Digital bundles with their 5-bay external bay.  So if you external bay supports all 5 drives with one cable, then this card will work.

Has anyone tested this with a port multiplier enclosure ?

I've used mine with 2 Sans Digital 4 disc SATA 3 enclosures for quite some time, One has 4 4 TB drives and the other an assortment of different drive sizes.  Works well. Transfer speed is restricted for simultaneous high volume copy loads between enclosures since it is a x1 lane card. But it works well for the media server and home backup duties for which I use it.

It does audible alarm you hot remove drives though which my previous eSATA controller didn't do. It may be configurable to turn that off, but I just sleep and re-wake the machine since I do it so seldom.

Question 4

Since eSATA has the option to plug a usb connector into it, can i attach a 10 port hub to this and still use all of the hdd's in the raid array?

This card has NO USB connectors of any kind. With two eSATA connectors, you could connect one RAID box to one of the connectors and I suppose some kind of eSATA to USB adapter to the other.

I have not used any such adapter on either of my RAID cards, but the two connections should be able to cooperate. However, whether those USB drives would be available to the RAID array might depend on the software. The latest web GUI from Highpoint is feature-rich, so it may work well. You should probably contact Highpoint with that question for a definitive answer.

My card did not come with a full height PCI bracket as indicated in the photo. I contacted Highpoint and TECH EXPERTS regarding this and they were no help. Has anybody had any luck finding a full height bracket?

Mine also came with both full height and half height brackets.  Beware of software however.  It comes with no drivers.  I purchased mine in Mar 2018.
The documentation included states that for Windows 7, Vista, 2008 download the driver from the highpoint site, however the newest OS driver for the 622 is for Server 2003/windows XP.  There is a 644 drive that will support windows 10, but it doesn't work on the 622.  The downloaded data sheet says that the Chip on the card is a Marvell 88SE9120 but there is no software on the Marvell Site for any raid product.  
I have used the 644 on server 2008 with a SansDigital Tower for a 20Gb Raid 5 Array (16 useable).  Sans Digital seems to be selling their enclosures without RAID cards now, so I thought I would pick up a cheap replacement card.  The description should indicate that this is a legacy product and not supported for current operating systems.

Question 6

Trouble trying to upgrade the BIOS to 1.1 on older PC. Can't boot from USB on this motherboard. I attached an external BluRay drive, but bootable DOS 6.xx CD cannot see the drive. Don't have an old external CD/DVD drive to boot from. Any suggestions? ABit Fatality FPIN9 motherboard.

you need to make a dos 6.xx floppy disk with bootable CD Drivers on the disk and specified in the config.sys  and MSCDEX specified in the autoexec for a CD that will boot and see itself.  Much easier to get  a bootable DOS 6.22 ISO to burn a Boot CD or USB flash drive that will boot up and see the cd drive at allbootdisks.  google it
