No, the key, when locked, simply keeps the drive from being removed.

Does this fit in a 5.25" drive bay?

It's a 3.5" device, so you'll need an adapter, but they are cheap; and quite a few cases include them in the accessories. The adapter attaches in the case like any 5.25" device, and just has some arms that fit this like any 3.5" bay. It'll also usually include a faceplate with a cutout for a clean appearance. That device, though, is built like a rock. It's extremely high quality, and well worth picking up an adapter. They're not expensive.

How come there are no screw holes on the bottom of this unit?

There's no standard bottom screw holes position for a 3.5" device that is why. The 3.5" device uses different screw hole position than 3.5" hard drive, even the side screw holes position is different. People thought that the 3.5" device and 3.5" hard drive are using the same screw holes position is not correct. They are different.

Will this rack enclosure affect the speed of my hard drives?

The rack enclosure supports SATA 6Gb/s with no speed reduction.
