Will these be compatible with the Klipsch K-100SW 10-Inch Powered Subwoofer?

Maybe a little late for an answer, but I think that combination sounds like it should be pretty nice. I just got a pair of these to use as front L-R channels with a Velodyne VX-11 powered 10" sub (crossover point set at 80Hz), and the combo sounds great to my ears.

Question 2

I just bought and installed a full set of R-10s in a 5.x setup.  Would these be a good upgrade for the front 4" RB10s?

Depends what you want. I use these as wides, not mains, and they are great for that. I listened to them as mains for a few minutes and they were surprisingly good  for being so small. They will be slightly louder than the ones you have, and go a little lower in the bass. Either way, you should have a subwoofer. You could re  purpose the old ones into a 7.x.
