can I use a reg 3.5" HD or must I use an SSD drive?

You can only use this with an msata SSD disk A regular 3.5" HD is out of thye quetion whether it is an SSD or not

Maybe, but I doubt it. This is an mSATA enclosure...not M.2, which is a different form factor.

Just upgraded my mid-2015 mac book pro 128GB SSD with a larger one. Will my old 128Gb PCIe M2 Samsung SSD fit and work?

This enclosure is for mSATA SSDs only, the SSDs that use the "mini PCIe" (and M.2) interface are different and not compatable with this enclosure. Just google "msata vs mini pcie ssd" to understand the differences between the different types of SSDs
