Why does this have a DC supply plug in? It doesn't come with a power supply and it won't work otherwise.

It works without DC power with most computers. However some machines will not provide enough current over USB, and it's only then that the supplemental DC power is needed.

Has it got LED?

There is a blue LED in the front.  It is on but dim to indicate it has power, and then flashes brighter to indicate activity.

Question 4

The unit has a DC input jack. Which adapter is for that? I don't see one listed on the page.

It does not come with an adapter, but I bet it uses the same 3.5mm 5v adapter that similar drives use. These also exist in USB to 3.5mm versions if you do not have a power outlet handy. You will only need the adapter if the USB port is unable to supple sufficient power. I'm currently using a 7200 RPM Seagate drive without needing additional power.

How do you use the OTB on this product?

One Touch Backup (OTB) needs the installation of a backup software/application that can take advantage of this OTB function.
You can install one from the CD that came in the package.
You have choice of AsMedia, GL830, Initio, Jmicron.
Read the instruction/system requirement in each respective PcCloneExLiteEng.chm file.
I have not utilized or used this OTB feature myself.

Question 6

Will it hold my 1.5 in, 4 in 6 in 1000GB hard drive?

You mean a 1TB drive with dimensions 4x6x1.5 inches?  That would be a 3.5" desktop drive and the answer is no.  This is meant for 2.5" laptop drives.  For reference, google search on wiki hard disk drive form factors
