WILL THIS WORK ON WINDOWS 10 O.S. ?

I'm not using Windows 10, but I believe the device should work with just about any O.S. as it is essentially just electrical/mechanical adapter and it requires no drivers or software of any kind.  It does require a standard 5.25" drive bay and it and probably won't work readily in cases with proprietary drive bay rail systems.  To be functional the USB ports require a motherboard with a 19-pin USB port or an appropriate adapter or add-in card.  Currently using fully featured in many old netburst and core 2 machines.

How do the 'doors' work on the 2.5" and 3.25" bay? 
Does the 3.25 function like old school VHS dust door?

There is a button that slides to the right to open the 2.5 door and a lever for the 3.5. Both swing open outward and to the right. On the inside of each door there is a spring that pushes on the back side of the drive to fully engage it onto the powers and data ports. Opening the door pulls a catch to release the drives. Because of this, drives will not function with their door open.

Can a 3.5" front panel hub fit inside the lower opening (instead of the 3.5" hard drive) to add more i/o?

Wouldn't have thought so, but I have a 3.5" USB front panel and it did slide into the opening meant for a 3.5" hard drive.  Would say mounting a 3.5" front panel may be a little bit of a challenge as there are no typical mounting holes on the sides or bottom.  There is a narrow bottom lip that you could possibly drill holes into to mount from the bottom.  Another thing to contend with would be the door which is 5/8" thick and has a spring clip attached that makes part of the door be 7/8" thick.  The door has linkage that causes a lever at the back to move frontwards when the door is opened (to push a hard drive out).  Leaving the door on would mean the 3.5" front panel would be recessed about 7/8" and if the front panel was not real long, would not interfere with the lever.  Removing the door may be a challenge.

Suggestion: Assuming you want to keep the removable 2.5" hard drive capability and/or the two USB 3.0 ports.  If you only want to mount a (non-removable) 2.5" hard drive and don't need the USB 3.0 ports, you may want to consider the "ICY DOCK Dual 2.5 SSD 1 x 3.5 HDD Device Bay to 5.25 Drive Bay Converter..." or look at some of the other ICY DOCK mounting adapters.
