Personally I have an 8 TB drive partition specifically for all of my games in Windows 10. 
There are only two requirements to have a partition larger than 2TB. The first is to use GUID Partition Table (GPT) partitioning scheme instead of the older Master Boot Record (MBR) partition scheme. Windows 10 does this by default I believe but if you're adding this as a secondary drive you'd need to make sure you select GPT instead of MBR when setting up the partitions. (The one review stating Win 10 doesn't support it probably set theirs up using MBR). The second requirement is only applicable if you are installing Windows onto the drive larger than 2TB, which is to use a Unified Extensible Firmware Interface (UEFI) and not BIOS interface when installing Windows. This is a motherboard and install medium setup. As long as you have an UEFI install drive and a motherboard that supports it you won't have any problems making a GPT partition larger than 2TB.

TL/DR: Yes as long as you set it up with GPT if a secondary drive or have a semi-recent setup that supports UEFI if making it your Windows install drive.
