Will a drive in the 2nd bay mount in the OS, or is it for hardware duplication via the dock only? I would like to be able to have two drives in the dock and have them both show up in my OS.  Thank you in advance for the help.  -- Bill

We use it to duplicate 2.5" SSD to 2.5" SSD and 2.5" SSD to M.2 SSD (with 2.5" SATA adapter).  Works great!  Just remember that it wants to copy Port 2 to Port 1 (seems backwards to me - but do it wrong once and you will remember!)

Will the StarTech USB 3.1 Standalone Duplicator with with mSATA drives?

I have only used it so far to read an mSATA drive - haven't tried duplicating one yet.  You will need an adapter of course. It read fine so I cannot see why it wouldn't duplicate as well.  Hope this helps!

With Windows 10, if the copy replaces the master for the same computer, will it start and resume updates? Will Microsoft recognize it as a clone and reject it? Will the computer recognize it as the same drive verbatim?

First, a disclaimer: I am not an expert in cloning bootable drives... but the general answer is that if you successfully clone a bootable drive, the clone (copy) will replace the original perfectly... Windows will continue to update... and all will continue without a hiccup. Here is the catch: I have not found it to be easy to clone drives and have the copy boot successfully. In fact, that was the reason I bought this Standalone Duplicator Dock; I hoped that using the built-in duplication function might succeed better than my other efforts. Alas, for me it did not. I've tried several disk clone utilities and it is easy to succeed when one is copying a data drive... but for me it has been hit-and-miss successfully cloning a boot drive and have the copy actually boot. So, here's the solution that works for me: I buy Samsung SSD's and download Samsung's "Data Migration Software" to clone the drive. I use this Dock to connect the new, blank drive to the computer with the drive I want to clone. That approach always works for me... the new copy made with the Samsung software always boots. (Note that the Samsung Data Migration Software ONLY works if at least one of the drives... either the original or the new copy... is a Samsung drive). Other folks might be able to tell you the "trick" to using this Dock to clone a drive and to make the clone bootable... but I've not been able to do it. I've decided that it is worth spending a little more money on new Samsung SSD's because I know I will be successful using them (and the Samsung Data Migration Software) to clone boot drives. Hope that helps!

I would like to use SSD that are 12TB or more, will it work in this dock?

My guess is the size of SSD doesn't matter with to the Dock.  It would really depend on your system BIOS and if it can support really large hard drives.  We've used hard drives up to 4 GB with this dock.  You must have some deep pockets to be using 12 GB SSD.

Why can't I create spanned or striped disks with this product?

I don't think that is the intended use of this device.   Sounds like you need a RAID array.
The startech is better suited to making a back up copy of your drive or conversion to SSD.
