Can I remove the drive from its enclosure and use it as an internal hard drive?

I have been able to remove the drive from these enclosures with great care and gentleness and not break the case. It is very easy to break the case. The internal drive is not guaranteed to be any specific model, but often a baracuda. In the U.S., you do not void warranty by opening and putting back a piece of electronics (although you are still likely to get a big hassle trying to do a warranty claim).

I'd recommend a thorough test of the new drive before shucking it. Badblocks takes about 5 days to check every sector, but it's easier than figuring out a problem after you've broken the drive down.

Sorry, I can't say with a 100% certainty that it would work that way.  A lot would depend on your router's handling of USB connected devices.  If a Windows formatted USB flash drive connected to your router automatically shows up as a network drive, this drive (it requires its own power source) should also theoretically show up the same way.
